Studying gamete biology can provide important information about a species fertilization strategy as well as their reproductive ecology. Currently, there is a lack of knowledge about how long sea bass Dicentrarchus labrax eggs can remain viable after being activated in seawater. The objectives of this study were to understand the effects of pre-incubation of fresh and overripe sea bass eggs in seawater and to determine the duration of egg receptivity. Pooled eggs (fresh and overripe) from four females were pre-incubated in seawater for 0 min (control), 0.5 min, 1 min, 3 min, 10 min and 30 min and then fertilized by pooled sperm from four males. The deliverable (D7.1) of WP7 “Establishment of a Computer Assisted Sperm Analysis (CASA) for the evaluation of grey mullet sperm” has been submitted. Preliminary analyses show that mullet sperm motility pattern is not similar to that of sea bass (Dicentrarchus labrax) or rainbow trout (Oncorhynchus mykiss) in terms of velocity as well as motility duration (ca 1 min; Fig. 1a). Our preliminary results, indicate that the major bio-technical settings, dilution of sperm, quality of activation and mastering of video recordings have been determined and can now be applied to experimental protocols. Androgenesis is a form of uniparental reproduction leading to progenies inheriting only the paternal set of chromosomes. It has been achieved with variable success in a number of freshwater species and can be attained by artificial fertilization of genetically inactivated eggs following exposure to gamma (gamma), X-ray or UV irradiation (haploid androgenesis) and by restoration of diploidy by suppression of mitosis using a pressure or thermal shock. The conditions for the genetic inactivation of the maternal genome in the European sea bass (Dicentrarchus labrax L.) were explored using different combinations of UV irradiation levels and durations. The sperm of seabass is very fragile and it quickly loses its ability to fertilize after collection either if kept undiluted or in classic saline media. In order to avoid cryopreservation when only short conservation is required, the process of sperm management including sperm collection, sperm dilution rate in storage medium and storage medium composition, was subject to experimental trials. A concentration of 20% urine generated a low pH of seminal fluid, and it immediately altered the motility ability. The term "domestication" has a legislative meaning when it applies to species submitted to selection pressures, but also a literary one, according to the time since which the species live closed to men and the possibility of controlling their reproduction. Bluefin tuna shows adaptative physiology features allowing life in a wide range of physical and climatic parameters like temperature, salinity and hydrostatic pressure, hence the species fulfil the main basic requirements for progressive domestication and rearing in captivity. The high mortalities recorded after handling and the losses observed after abrupt variations of environment in the cages, emphasised the extreme sensitivity to stress of tuna. As consequences, the progresses about the control of tuna biological cycle are very slow due to limited handling possibilities and tuna aquaculture is restricted to fattening of wild fish. At this time, bluefin tuna production progresses without domestication of the species. |

This work assesses the present knowledge on Pacific oyster sperm biology in comparison to two marine fish species (turbot and seabass) whose sperm characteristics are well described. Sperm morphology mainly differs by the presence of an acrosome in Pacific oyster which is absent in both fish species. In turbot as in Pacific oyster, a sperm maturation process along the genital tract is observed. Sperm motility is triggered by changes in osmolality for seabass and turbot and in pH for Pacific oyster. However, complementary factors are involved to maintain sperm immotile in the genital tract. After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization. |
